Repressive effect of miRNA-363 via SOX4 on human osteosarcoma cell growth and apoptosis

AIM: To detect the expression of miRNA-363 and SOX4 in osteosarcoma tissues and to investigate the effect of miRNA-363 on the viability and apoptosis of human osteosarcoma cell line MG-63.METHODS: Real-time PCR was used to detect the expression level and the relationship of miRNA-363 and SOX4 mRNA in the osteosarcoma tissues and the corresponding paratumorous tissues collected from 63 patients. The expression levels of miRNA-363 and SOX4 in osteosarcoma cell line MG-63 after transfected with miRNA-363 mimics were measured. The cell viability was measured by CCK-8 assay. Flow cytometry was used to monitor the changes of cell cycle and apoptosis. The changes of SOX4 and miRNA-363 expression levels in the MG-63 cells after transfection with SOX4 siRNA or pcDNA/SOX4 was detect by real-time PCR.RESULTS: The expressed level of miRNA-363 was lower, and the expression level of SOX4 was higher in the osteosarcoma tissues than those in the adjacent normal tissues. A significantly negative correlation between the expression levels of miRNA-363 and SOX4 was observed. The expression of miRNA-363 in the MG-63 cells after transfection with miRNA-363 mimics was significantly up-regulated, while the expression of SOX4 in the MG-63 cells was significantly down-regulated, with significant difference as compared with the cells transfected with miRNA-NC and control cells. The viability of MG-63 cells was inhibited, the cell cycle was arrested in G₀/G₁ phase, and the cell apoptosis was increased by transfection with miRNA-363 mimics. The relative protein expression levels of SOX4 in SOX4 siRNA group and pcNDA/SOX4 group were significantly different from those in negative control group, but the relative expression levels of miRNA-363 had no significant difference. Over-expression of SOX4 restored the viability of the MG-63 cells reduced by miR-363.CONCLUSION: The expression level of miRNA-363 is low in human osteosarcoma tissue. miRNA-363 may inhibits the viability of osteosarcoma cell line MG-63 and promotes cell apoptosis in vitro via inhibiting the SOX4 expression.     

Determination of immunochemically nonreactive urinary albumin by HPLC method and its significance of diagnose on children diabetic nephropathy

AIM: To detect the immunochemically nonreactive urinary albumin and its significance of diagnose on diabetic nephropathy by high performance liquid chromatography (HPLC).METHODS: Urine samples of 58 diabetic children were collected. Urinary albumin was measured by HPLC and by immunoassay respectively.RESULTS: The retention times of albumin of standard and urine samples were 14.3 min. The content of albumin by HPLC and by immunoassay were 9.6-195.1 mg/L and 0-158.6 mg/L, respectively.The level of urinary albumin was higher by HPLC detection than that by immunoassay (P<0.05) in the same sample. The microalbuminuria rates were 70.7% detected by HPLC and 37.9% by immunoassay.CONCLUSION: Total urinary albumin can be measured by HPLC. Microalbuminuria in diabetic children can be observed earlier by HPLC.     

香石竹快速繁殖技术研究

以中花散枝香石竹"红芭芭拉"的茎段为外植体,探讨不同培养基对香石竹的诱芽萌动、继代增殖和生根培养的影响。结果表明:外植体切取部位的诱导芽萌动率从高至低排序为:顶芽带腋芽的茎段不带腋芽的茎段;当培养基为MS+6-BA 1.0mg/L时,对香石竹带腋芽的茎段芽萌动最好;在培养基为1/2MS+6-BA 1.0mg/L+NAA 0.2mg/L时,继代增殖长芽的效果最好;在培养基为1/2MS+6-BA 0.2mg/L+NAA 0.5mg/L时,诱导不定根效果最好。

ZnO/Na 2 O(mol)对cBN陶瓷砂轮性能的影响

通过DSC -TG综合热分析、X衍射分析、线膨胀测试仪、扫描电镜等测试方法研究了ZnO/Na 2 O(mol)（ Z 值）对Li 2 O -K 2 O -SiO 2 -Al 2 O 3 -B 2 O 3 系陶瓷结合剂及立方氮化硼砂轮性能的影响，结果发现：ZnO能够抑制α -石英晶相析出，诱导析出低膨胀晶体Li 2 OAl 2 O 3 7.5SiO 2 ，并随着 Z 值的增加，α -石英减少，Li 2 OAl 2 O 3 7.5SiO 2 晶体逐渐增多， Li 2 OAl 2 O 3 7.5SiO 2 晶型结构趋于完整，晶粒尺寸减小，结构更加致密；当砂轮配方一定时，砂轮的体积密度、抗弯曲强度、洛氏硬度均随着 Z 值的增加而增大，气孔率减小，磨削（干磨）铸铁（QT400）时磨耗比先增加后减小；当 Z =0.75时，结合剂的膨胀系数为6.902 9×10 -6 K - 1 ，耐火度为805 ℃，砂轮的烧结温度为845 ℃，抗弯曲强度为73.97 MPa，气孔率为15.65%，磨耗比最大为156.42%.     

酸樱桃新品种‘玫蕾’

 ‘玫蕾’是从野生酸樱桃实生选种而来。单果质量5.3 g，总糖含量8.92%，总酸度1.49%，可溶性蛋白质含量1.46%，维生素C含量0.152 mg · g^-1 FW，铁含量0.040 mg · g^-1 FW，钙含量0.914 mg · g^-1FW。乔木，6年树冠高达3.6 m，果实表面颜色为紫红色，有光泽，果实甜酸，酸味浓郁，汁多，果肉深红色。适宜陕西关中地区栽植。

桂西北地区杉木冰冻雪灾与恢复措施

2008年1月中旬以来我国南方大部分省区遭受了历史罕见的雨雪冰冻灾害,使广西的森林资源和林业基础设施遭受惨重损失。针对广西西北部地区杉木(Cunninghamia laceolata)的受害类型和严重程度,结合杉木的生长特性和实际生产需要,提出相应的恢复技术。     

Effects of cytokines on renovation of acute renal failure in mouse with bone marrow derived mesenchymal stem cell transplantation

AIM: To observe the effects of cytokines on renovation of acute renal failure (ARF) in mouse with bone marrow derived mesenchymal stem cell (MSC) transplantation. METHODS: ARF animal model was induced in mouse by subcutaneous injection of cisplatin. Mice were randomly assigned into 3 groups: normal control group, ARF group and MSC group. After 24 h cisplatin injection, animals were injected intravenously with MSC in MSC group. Animals were sacrificed at 1 d, 4 d, 7 d, 14 d and 28 d after cisplatin injection. The blood urea nitrogen (BUN) and serum creatinine (Scr) were measured. The renal morphologic changes were scored with Paller’s criterion on hematoxylin and eosin (HE) stained sections. The mRNA and protein expressions of HGF, BMP-7, TNF-α and IL-10 were detected by RT-PCR and immunohistochemistry method. RESULTS: After 4 d of cisplatin injection, the BUN and Scr values in MSC group were significantly lower than those in ARF group (P<0.01). After 7 d and 14 d, the values of BUN and Scr in MSC group were still lower than those in ARF group (P<0.01, P<0.05). The renal morphologic scores of MSC group were also lower than those of ARF group. After 7 d, the expressions of HGF, BMP-7 and IL-10 were higher in MSC group than those in ARF group, the expression of TNF-α in MSC group was lower than that in ARF group. CONCLUSION: MSC promotes the recovery of acute renal failure induced by cisplatin. The mechanism may partly depend on paracrine of growth factor and amelioration of inflammatory.
